Abstract

Background: Analysis of human reports and comparison with results of exper-imental animals indicate that the effects of progesterone on human not analo-gous to experimental animals fetus, many studies showed that exposure to pro-gesterone during developing of genital tract of human fetus was not teratogenic. Other studies which performed on laboratory animals found association between progesterone administration during gestation and genital malformation. Objectives: to explore the effect of progesterone in 10.2mg/kg intraperitoneal injection in mice on testis development and anogenital distance. Materials and Methods: ten pregnant mice divided into five mouse control group that injected10.2mg/kg sesame oil and treated group that injected proges-terone at dose 10.2mg/kg/day allover gestation to the seventh day postnatally, mice offspring sacrificed, the anogenital distance, weight and diameter of testis were measured ,histological slides were prepared and examined for histopatho-logical change and measurement of seminiferous tubules diameter, germinal epithelium thickness and interstitial space were done . Results: the current study found anogenital distance decrement and histopatho-logical changes in mice offspring testis as hemorrhage or congestion of blood vessels ,irregular shapes of seminiferous tubules with atrophy, detachment of spermatogenic cells from basement membrane, death of spermatogenic cells and destruction of Sertoli and leydig cells ,it is also showed wider interstitial tissue and thinner germinal epithelium. Conclusions: The present study indicate study that progesterone injected in mice at dose 10.2 mg/kg /day (the equivalent human dose allover gestation was em-bryotoxic and teratogenic, it may cause feminization of male offsprings repre-sented by decrement the anogenital distance in mice and hypospadias in human .

Abstract

Amino acids have the role in the process of proteins synthesis. They are an essential source of nitrogen atoms that have a role in the pathways of synthetic reaction pathways. The carbon skeletons of the amino acids are the source of energy in addition to their role as precursors in the paths of interactions. The amino acids analysis for the brain of the quail bird in different stage of de-velopment (10-16 days of incubation) in addition to the hatching stage (17th day) and the adult. Materials and Methods: Amino Acids Analysis The amino acids were separated from the embryos and adult brains of the quail bird Coturnix coturnix and were diagnosed based on standard amino acids, using high perfor-mance liquid chromatographic device (H.P.L.C.). Results: The results revealed that there are 16 amino acid: Aspartic acid, Glutamine, Ser-ine, Histidine, Threonine, Glycine, Arginine, Alanine, Tyrosine, Cysteine, Va-linine, Methionine, Isoleucine, Phenylalanine , Lysine, and the highest concen-tration of amino acid at age of [(13,13) days of incubation, adult, (13, 13, 16) days of incubation, adult, (13) days of incubation, adult, (13, 13, 11, 13, 13) days of incubation, adult, (13) days of incubation] respectively and the low concen-tration at age of [(14, 12, 10, 10,12,10,10,10,10, 14, 12, 12, 15, 10, 14, 10) days of incubation respectively]. Conclusions: It concluded from this that there was found 16 amino acid that were analyzed in the brains of the quail embryos at the stage (10-16) days of incubation and hatching stage (17) days of incubation and adult (Asp, Glu, Ser, His, Thr, Gly, Arg, Aln, Tyr, Cys, Val, Met, Ili, Leu, Phe, Lys, and Lys) the highest concentration of aging (13, 13, adult, 13, 13, 16, adult, 13, 13, 11, 13, 13, adult and 13) respectively, and less concentrated in ages (14, 12, 10, 10, 10, 10, 14, 12, 12, 15 10, 14, 10) days of incubation, respectively.

Abstract

Background: Zygote produce from once a sperm fertilizes an egg cell. Then, the zygote (unicellular) will begin chain of cellular cleavages to produce multicellular mass, its embryo, the differentiated to different tissues and organism. The development of the embryo is called embryogenesis. Coenzyme Q10, is an antioxidant produced in the body. It boosts cellular energy and may enhance the immune system. CoQ10 is present and measurable in seminal fluid, the concentration of CoQ10 directly correlates with both sperm count and motility. It is beneficial in the prevention and treatment a wide range of health problems. Objectives: The present study was aimed to investigate the possibility of using coenzyme Q10 to improvein vitro fertilization (IVF), and early embryonic development (ED) in mice as a model for human being. Methods: Superovulation program was achieved to mature healthy female mice with age 10-12 weeks and weight 24-26 gm. After sacrificing female, oocytes were collected and incubated within CO2 incubator for less than 1 hour. Sperm were collected from vas deference of males. Sperm parameters were assessed after 30 min. ofincubation. Mature oocytes were divided into three groups according to theconcentrations of CoQ10 including G1 (control group; SMART medium only), G2 (treated group; SMART medium enriched with 20 M CoQ10) and G3 (treated group; SMART medium enriched with 40 M CoQ10). IVF technique was performed for 3 groups, and assessment of IVF (%), embryonic development stage (%) and abnormal embryo morphology (%) for each embryo stage. Results: Results of the present study appeared significant increment (P<0.05) in the percentages of IVF for both treated groups as compared to the control groups. Also, significant increase (P<0.0P) in the IVF (%) was observed when using 40 M CoQ10 as compared to 20 M CoQ10. Non significant differences (P>0.05%) in the 8-cells embryo stage were assessed among control and treated groups. Conclusion: From the results of the present study it wasconcluded that the coenzyme Q10 (40µM) enriched to the culture medium improved percentage of in vitro fertilization and no effect on embryonic development.

Abstract

Node-like receptor pyrin 3 is a tripartite protein composed of an amino-terminal pyrin domin (PYD), nucleotide binding NACHT domine, and a carboxy-terminal leucine-rich repeat (LRR) domine, and consider an intracellular sensor that detects a broad range signal and resulting in the formation and activation the NLRP3 inflammasome. This complex lead to the release of the inflammatory cytokines IL-1β, IL-18 which is important for host defense against invading pathogen. Modern evidence indicates that the NLRP3 inflammasome is involved in gestational hypertension. This study was conducted on fifty pregnant women and divided into two groups: 25 placentas from normotensive mothers having no hypertension before as the control group and 25 from gestational hypertension women as gestational hypertension group, obtained from the Department of Obstetrics and Gynecology in Almosil El Aam Hospital and Alkhanssa Teaching Hospital, Mosul city, Iraq. The placental tissue samples were collected from the central region and processed for an immunohistochemical technique to t h e expression of NLRP3 in placental tissue. The immunohistochemical study of t h e placenta of the gestational hypertension group showed the localization of NLRP3 protein and staining intensity in trophoblast, stroma, and endothelium villi were high quantification compared with placenta from the control group.

Abstract

Gestational diabetes mellitus (GDM) is a serious pregnancy complication in which a woman who has never had diabetes develops chronic hyperglycemia during her pregnancy. Normal placental function is essential for optimal fetal growth. The transport of glucose from the mother to the fetus is critical for fetal nutrient demands and can be stored as glycogen in the placenta. However, the function of this glycogen deposition is unknown: It may well be a source of fuel for a placenta itself or the storage reservoir for the later use by the fetus in times of need. While the significance of the placental glycogen remains unknown, the mounting evidence indicates that the altered glycogen metabolism and/or deposition is associated with many pregnancy complications, such as gestational diabetes, that adversely affect fetal development. The aim of this study is to assess glycogen deposition using Histochemical staining of Periodic Acid Schiff (PAS) stain. The placenta tissue collected from 50 women were enrolled in this study (25 women with no complications) and (25 women with gestational diabetes). The placentas of the two groups were compared in this study based on glycogen deposition with periodic acid-Schiff stain. The results of a histochemical investigation using PAS stain revealed a significant increase in the glycogen deposition (p≤0.001) in diabetic women's placentas within the intervillous core, around fetal vessels, and the basement membranes.
