Abstract

Lead is a heavy metal toxicant, present widely in the environment and workplaces. Zinc has been shown to reduce the toxicity of lead. The present experimental animal study was designed to observe the changes in the testes due to excessive use of lead. This study aims to detect the effect of antioxidants (zinc) on the toxicity of high doses of lead on the reproductive organs of male mice embryos compared to the control group as a model for human representative. Thirty mature female mice were used (Swiss-Webster), as a mammalian model. They were divided into three groups, 10 mice/group after labelling them. They were randomly assigned into three experimental groups, and these embryos were processed for histological observations. The recorded results showed a severe disruption in the organization of testicular structure with primordial germ cells severely degenerated with the absence of basal lamina and peritubular myoid cells of the treated group (G1) compared with the control group at day 14 and day 17. The embryo testes of (G2) at day 14 showed irregular testes cords with prominent degenerative changes compared to that of the control group (G3). It was concluded from these results that zinc sulphate plays a significant role in the improvement of histological damage of the testes belonging to mothers injected with lead acetate and protects the reproductive organs from injury during lead toxicity.

Abstract

The toxic effect of drugs and environmental chemicals on the human reproductive system has become a major health concern. Lead is recognized as the most dangerous of the toxic metals and a major threat to the biological systems of both human and animals. In this study we detect the effect of two types of antioxidants: flaxseed and zinc, on the toxicity of low dose of lead acetate on reproductive organs of male mice, this study was conduct. Two groups of mature male mice, exposed to low dose of lead, were administered either zinc sulphate (0. 5mg/kg body weight/day) or flaxseed oil (2.5% of total daily food intake), for 6 weeks. The results showed that lead caused a significant reduction in the total body weight, testicular weights & adverse effects on sperm parameters, while the concomitant administration of antioxidants caused elevation in these weights with significant improvement in sperm quality as compared to the only lead treated group. It is concluded that lead, like all other heavy metals, could induce oxidative stress and the concomitant administration of antioxidants (zinc or flaxseed) can ameliorate the toxic effect of lead on reproduction.

Abstract

Background: The effects of Lead and other essential metal like zinc and copper on the reproductive system have been previously reviewed. Objective: Our goal in this study was to estimate the levels of Lead, Zinc and Copper in patients with polycystic ovary syndrome and to confirm the effect of such metals on the female reproductive system at environmental exposure. Materials and Method: Fifty two women in their reproductive age (20-40) years old, which had been diagnosed as PCOS. Venous blood sample were taken from each patient and control, blood lead, zinc and copper were determined by atomic absorption spectrophotometer. Hormone assay were determined by ELISA (Enzyme Linked Immune Sorbet Assay) technique. Results: This study showed a significant increase of blood lead and serum zinc, copper (p<0.01), in addition to testosterone, lutenizing hormone, LH/FSH ratio and body mass index (BMI) (p<0.05) in patients with polycystic ovary syndrome when compared with control. Conclusions: On the basis of the observations of the present study, it can conclude that the increased levels of Lead as a result of the exposure to this metal may be a risk factor for the development of polycystic ovary syndrome in women and has adverse effects on female endocrine and reproductive function.

Abstract

Pregnancy-Associated Plasma Protein (PAPP-A) is a zinc metalloproteinase in the insulin growth factor system (IGFs) produced by the syncytiotrophoplast region of the placenta. It plays a critical function in the cleavage of IGFBP4. In the ovary IGFs, it regulates follicular and oocyte maturation, and steroidogenesis. While in polycystic ovarian syndrome (PCOS) Hyperinsulinemia and hyperandrogenemia it causes follicular environment changes and early ovulation resulting in lower oocyte and embryo quality in patients and this will decrease the success of pregnancy in women enrolled in the ICSI cycle. The present study aimed to assess the relationship of PAPP-A levels in serum and follicular fluid in women with PCOS and non-PCOS with oocyte and embryo quality in women undergoing ICSI cycle. 45 infertile Iraqi women were enrolled. Women with PCOS had to meet at least two of the three criteria set by the Rotterdam ESHRE/ASRMS criteria, the age of the included women ranged between 20-45 years. In non-PCOS patients, PAPP-A has higher level in serum and follicular fluid but without a statistically significant difference matching with PCOS group. In addition, there was no significant correlation between PAPP-A levels in serum and follicular fluid with oocytes and embryo characteristics. However, PAPP-A levels are higher in serum and follicular fluid in women with positive pregnancy but without significant differences. PAPP-A had no correlation with oocyte and embryo quality.
